Catalyzes the conversion of L-tyrosine to L-dihydroxyphenylalanine (L-Dopa), the rate-limiting step in the biosynthesis of cathecolamines, dopamine, noradrenaline, and adrenaline. Uses tetrahydrobiopterin and molecular oxygen to convert tyrosine to L-Dopa (By similarity). In addition to tyrosine, is able to catalyze the hydroxylation of phenylalanine and tryptophan but with lower specificity . Positively regulates the regression of retinal hyaloid vessels during postnatal development (By similarity). .

Background
Tyrosine hydroxylaseortyrosine 3-monooxygenaseis theenzymeresponsible for catalyzing the conversion of theamino acidL-tyrosinetoL-3,4-dihydroxyphenylalanine(L-DOPA).It does so usingmolecular oxygen(O2), as well asiron(Fe2+) andtetrahydrobiopterinascofactors.L-DOPA is a precursor fordopamine, which, in turn, is a precursor for the important neurotransmittersnorepinephrine(noradrenaline) andepinephrine(adrenaline). Tyrosine hydroxylase catalyzes the rate limiting step in this synthesis ofcatecholamines. In humans, tyrosine hydroxylase is encoded by theTHgene,and the enzyme is present in thecentral nervous system(CNS), peripheral sympathetic neurons and theadrenal medulla.Tyrosine hydroxylase,phenylalanine hydroxylaseandtryptophan hydroxylasetogether make up the family ofaromatic amino acid hydroxylases(AAAHs).
