Description

Involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) and O4-methylthymine (O4-MeT) in DNA. Repairs the methylated nucleobase in DNA by stoichiometrically transferring the methyl group to a cysteine residue in the enzyme. This is a suicide reaction: the enzyme is irreversibly inactivated.

	Western blot analysis of extracts(35ug) from 9 different cell lines by using anti-MGMT monoclonal antibody(Clone UMAB56).
	HEK293T cells were transfected with the pCMV6-ENTRY control(Left lane) or pCMV6-ENTRY MGMT(Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates(5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-MGMT(1:500).

Immunofluorescent staining of HeLa cells using anti-MGMT mouse monoclonal antibody(MG823463,green,1:50). Actin filaments were labeled with Alexa Fluor® 594 Phalloidin(red),and nuclear with DAPI(blue).
